Synthetic cells hold great promise in the development of innovative therapeutic strategies. However, molecular payloads synthesised inside synthetic cells must be secreted and new molecular building blocks or drugs must be delivered to synthetic cells in a smart and programmable way. Condensates have recently been shown to interact with membranes in a rich and dynamic way, including the possibility of entering  giant unilamellar vesicles (GUVs) via endocytosis. In this project, you will develop synthetic cells and condensates that exhibit a tunable affinity for the GUV inner membrane. The membrane affinity will be controlled via biochemical pathways that regulate ATP levels and that regulate the self-organisation of protein filaments. Enzyme-controlled condensates will be directed to the membrane, where condensate property modulation and membrane remodelling are used to directly uptake or  secrete condensates through membrane wrapping to deliver biomolecular payloads concentrated inside the condensates. This may enable future use of synthetic cells for the development of RNA and protein-based therapeutics.

Traditional spray drying relies on mechanical atomization to create droplets and uses hot air to evaporate the solvent. This process often requires higher temperatures, which can lead to the degradation of thermosensitive and air-sensitive compounds. Additionally, traditional spray drying may produce a broad particle size distribution and poor control over particle morphology. On the other hand, ESD is an advanced drying technology that utilizes electrostatic forces to produce ultra-fine, uniformly sized particles at lower temperatures under inert atmosphere. In ESD, an electrical charge is applied to the liquid feed, causing it to disperse into fine, charged droplets under the influence of an electric field. These droplets have a high surface area-to-volume ratio, allowing for rapid solvent evaporation and drying at significantly reduced temperatures compared to conventional methods. Furthermore, ESD is environmentally and economically advantageous compared to other drying techniques because it consumes less electricity and operates at lower temperatures, leading to reduced energy costs and a smaller environmental footprint. In alignment with sustainability goals and circular economy principles, this project will also explore the use of encapsulating materials derived from waste streams, such as agricultural by-products or food industry residues. The goal of this doctoral project is to develop advanced electrostatic spray drying processes to produce powders encapsulating thermosensitive compounds. You will investigate the optimization of process parameters, explore different encapsulating materials from waste residues, and assess the stability and release profiles of the encapsulated compounds under various conditions.

In the B-ACTIVE network, fundamental and clinical scientists and companies will study the impact of posttranslational modifications of chemokine ligands and receptors and train 10 doctoral candidates (DCs) to future experts in this domain with a translational and interdisciplinary mindset. The DCs will characterize interactions between chemokine ligands, receptors and glycosaminoglycans, study signaling pathways and recognize their importance in inflammatory reactions. DC10: We are seeking a highly motivated and intellectually curious PhD candidate to contribute to this project at Copenhagen Center for Glycomics (CCG), University of Copenhagen, which is a world leader in genetic glycoengineering and dissection of biological functions of glycans. CCG has state-of-the-art facilities for mass spectrometry, mass photometry, flow cytometry, cell engineering, and glycan arrays, and CCGs group leaders hold diverse expertise in all areas of glycosciences. Our core resources include glycoproteome data and bioinformatics tools, validated gene editing tools, glycoengineered cell libraries, and design matrices for the recombinant production of glycotherapeutics. The overall aim of the network is to advance cancer treatments by acquiring a new understanding of the earliest macrophage-tumour triggered tissue changes at the metastatic site, leading to better response prediction and more effective (immune) treatment options for cancer patients. The project will empower scientists, physicians, health care professionals, pharmaceutical industry, policy makers, patients and the general public to embrace the novel concept of selecting patients for personalized and improved (pan)cancer immunotherapy options based on the metastatic site. Ultimately, the project will support medical doctors and health care professionals to develop new guidelines for the improved and simplified application of “metastasis immunotherapy” throughout Europe.Collaborating with other Doctoral Candidates of the network, you will establish highly innovative preclinical Organ-on-a-Chip (OoC) models to faithfully mimic the very early events of tumour cell invasion and macrophage interactions in the context of healthy OoC models. Your tasks will be to establish standardized human bone OoC integrated with (age-relevant) blood monocyte-derived macrophage population, to identify macrophage and matrix changes induced upon interaction with BCa and PCa cells in a bone OoC model and to select and test potential macrophage-targeted compounds to interfere with tumour cell survival and invasion into bone OoC. Finally, you will use your findings to write and defend your thesis to obtain a doctoral degree.

PREDICTOR aims to establish a rapid, high-throughput method to identify and develop materials for electrochemical energy storage. It will enable the rapid identification, synthesis and characterization of materials within a coherent development chain, replacing conventional trial-and-error developments. To validate the PREDICTOR system, the case study will be active materials and electrolytes for redox-flow batteries. Within the project, three demonstrator battery cells (TRL3-4) will be assembled and tested with the newly developed materials.
Project objectives:
•  A modelling and simulation tool for the computational screening of organic chemicals based on their potential performance in energy storage systems.
•  Automated chemical synthesis, electrolyte production and characterization methods, so that the chemicals identified in the screening step can be rapidly produced and tested for their suitability in energy storage applications.
•  Artificial- intelligence- based self-optimization methods that allow experimental data from material characterization to be fed back into automated experimental methods to enable self-driving laboratory platforms and for modelling and simulation tools, improving their accuracy.
