पुनः संयोजक डीएनए प्रौद्योगिकी के चरण
(1) दाता जीव से उपयोगी डीएनए खंड को अलग करना।
(2) यह सुनिश्चित करने के लिए कि प्रत्येक वेक्टर को एक से अधिक डीएनए टुकड़ा प्राप्त नहीं होता है, इसे एक उपयुक्त वेक्टर में विभाजित करना।
(3) उसके पुनः संयोजक डीएनए की कई प्रतियों का निर्माण।
(4) इस परिवर्तित डीएनए को प्राप्तकर्ता जीव में डालना।
(5) रूपांतरित कोशिकाओं की स्क्रीनिंग।
वेक्टर:
जेनेटिक इंजीनियरिंग में वेक्टर आमतौर पर एक डीएनए खंड होता है जिसे चयनित डीएनए को जीवित कोशिकाओं में स्थानांतरित करने के लिए वाहक के रूप में उपयोग किया जाता है। ये इस प्रकार हैं:
(1) प्लास्मिड: प्लास्मिड अधिकांश यूकेरियोट्स में मौजूद डीएनए के अतिरिक्त क्रोमोसोमल, बंद गोलाकार दोहरे फंसे हुए अणु हैं। सभी प्लास्मिड डीएनए के प्रतिकृति टुकड़े ले जाते हैं जिनमें अनुवांशिक जानकारी होती है जिसे दोहराने के लिए आवश्यक होता है। प्लास्मिड पीबीआर 322 पहले व्यापक रूप से इस्तेमाल किए जाने वाले क्लोनिंग वैक्टर में से एक था, इसमें एम्पीसिलीन और टेट्रासाइक्लिन प्रतिरोध जीन दोनों होते हैं।
(2) फेज: यह फेज l क्रोमोसोम से निर्मित होता है और बैक्टीरियोफेज क्लोनिंग वैक्टर के रूप में कार्य करता है।
(3) कॉस्मिड: प्लास्मिड और फेज l क्रोमोसोम के बीच के संकर कॉस्मिड वैक्टर को जन्म देते हैं।
(4) इन सबके अलावा कृत्रिम गुणसूत्र होते हैं जैसे
(i) बीएसी (बैक्टीरियल कृत्रिम गुणसूत्र)
(ii) वाईएसी (खमीर कृत्रिम गुणसूत्र)
(iii) मैक (स्तनधारी कृत्रिम गुणसूत्र) यूकेरियोटिक जीन स्थानान्तरण के लिए बहुत कुशल वैक्टर हैं।
पुनः संयोजक डीएनए प्रौद्योगिकी का अनुप्रयोग:
पुनः संयोजक डीएनए की तकनीक को निम्नलिखित तरीकों से नियोजित किया जा सकता है।
(1) इसका उपयोग जैविक प्रक्रिया में आणविक घटनाओं जैसे सेलुलर भेदभाव और उम्र बढ़ने को स्पष्ट करने के लिए किया जा सकता है। इसका उपयोग सटीक जीन मानचित्र बनाने के लिए किया जा सकता है।
